Guitar Hero 5, esattamente come il suo predecessore Guitar Hero World Tour, proporr una tracklist di canzoni da suonare con i controller-strumenti (venduti separatamente) che replicano i quattro strumenti principali di una band, ovvero chitarra, basso, batteria e microfono per il canto, ma presenter diverse novit: tanto per cominciare le canzoni della tracklist non saranno pi da sbloccare nell'arco del gioco tramite un'apposita carriera ma saranno tutte disponibili fin dall'avvio della prima partita. In secondo luogo si segnala la possibilit di suonare nella nuova modalit "Party" nella quale i giocatori potranno "entrare e uscire" con il loro strumento a proprio piacimento durante l'esecuzione del brano stesso senza che questo costituisca una penalit come avveniva nei vecchi capitoli della serie. Un'altra aggiunta importante la possibilit effettuare sessioni multiplayer con i propri amici suonando tutti lo stesso strumento; infine c' da segnalare un'interessante feature che viene ripresa dal precedente capitolo che la compatibilit di Guitar Hero 5 con il negozio online unificato della serie che gi permette di acquistare le canzoni indipendentemente che si possegga Guitar Hero III: Legends of Rock o Guitar Hero World Tour.La tracklist sar composta da 85 canzoni di 30 artisti diversi e saranno tutte in versione originale. Tra i brani finora annunciati ci sono, tra gli altri: 2 Minutes to Midnight - Iron Maiden, All Along the Watchtower - Bob Dylan, No One to Depend On - Santana, The Rock Show - Blink 182, Sympathy for the Devil - Rolling Stones e Under Pressure - Queen e David Bowie. Saranno presenti anche artisti del calibro di Bon Jovi, Deep Purple, Duran Duran, Kiss e i Nirvana.
